Investigation of yeast properties during the beer fermentation

In this diploma thesis, the properities of brewery’s yeasts during the operation beer fermentation in the selected brewery and dependences of diferent agents and their influence on viability and vitality of the yeasts were studied. Other observed properities were physiological state, pH, temperature, a value of soft and coarse sludges, a value of dissolved oxygen in wort, the level of wort’s fermentation. It were collected the samples of fermenting wort and washing yeasts for determination. The used yeasts were observed from the first time until thein last used (generally 4 times). pH, temperature, apparent fermenting and total numer of yeast cells were measured in the collected samples. The used yeasts were washed before every other application and vitality was determined. The number of death cells were observed by this method. The number of death cells never fell bellow 5 %. The value of dissolved oxygen in wort is also the important agent for the growth of yeasts. The touchs with low number of oxygen showed that the time for fermentation had to be longer (by about 3 or 4 days more). The touchs with higher number of sludges showed that the sludges haven’t any influence on the yeasts and the procces of fermentation. Confirm the accuracy of the repeated used of the yeasts was the object of this thesis.
Vitality in the architectonic environment

More and more research studies are being conducted on the quality of architectural space as a factor affecting our mental, social and physical health. In the past, this topic was often overlooked in its complexity. The establishment of the specialized fields of building biology, biophilic architecture and the works of Christopher Alexander and Nikos Salingaros now allow us to consider various facets of this subject. This dissertation aims to analyse the topic of vitality in architectural space by studying the abilities and perceptions of its users. Therefore its aim is not to develop a new theory of “vitality-enhancing architectural space”, but to describe how this phenomenon is perceived by the users themselves. Users can provide feedback on which features of architectural space they see as the most important for the feeling of vitality. These features have been hierarchically organized based on their perceived importance, analysed and described in detail to allow their relatively easy application in practice. This dissertation will also allow a comparison with other existing works on the studied topic.
Examination of degradation frequently repaired shearing and forming tools

The diploma thesis is focused on the renovation of forming tools made of tool steel, their weldability and assessment of the quality of tool repairs. The damaged tool was a part of the cutter from the progressive cutting tool made of Caldie steel. We used micro TIG technology to repair the damage. A total of three types of additional materials were used and investigated to restore the geometry of the damaged edge. One of the main purposes was to evaluate the possibility of extending the life of tools and reducing the implementation of already inefficient repairs. A number of destructive tests were performed for the assessment - macrostructure, microstructure, hardness measurement and EDS analysis.
Lifecycle of selected building

In this thesis, the main objective is to determine the price of selected construction project during its life cycle and assess operational costs. As a selected object is a family town house built in 1982 as a building type okal. The theoretical part is devoted to professional concepts and detailed information regarding the lifecycle, durability, budget, and wear-related issues. The practical part includes an itemized budget and the actual award of a selected object by using the software on valuation. The resulting values of individual valuation methods are then compared and graphically displayed. Furthermore I judge the actual operating costs incurred during the period.
Reakce sazenic vybraných druhů dřevin na zaplavení

The thesis investigates the response to flooding of five species types of trees occurring in floodplain forests of South Moravia. This work aimed to evauate different lengths of the seedling flooding and examined seedling vitality and mortality. The investigated tree species were summer oak (Quercus robur L.), hornbeam elm (Ulmus minor Mill.), heart linden (Tilia cordata Mill.), hornbeam (Carpinus betulus L.) and field maple (Acer campestre L.). The flooding experiment was conducted from April to July 2022 and at different time intervals (7-14 days) the seedlings were flooded up to the root collar. Control seedlings were not flooded but only watered. 140 seedlings of each species were flooded and were gradually removed from the flood. The results show that relatively low mortality was observed for the flooded trees in all intervals. It was lowest for Quercus robur and Tilia cordata, which were characterized by high vitality in addition to low mortality. Ulmus minor was characterized by moderately vital seedlings. Acer campestre and Carpinus betulus had the highest mortality and their vitality decreased significantly towards the end of the experiment. The hypothesis of Quercus robur's resistance to flooding has been confirmed, which means the seedlings of this tree can probably cope well with flooding even in the conditions of floodplain forests. Tilia cordata also coped very well with flooding. It has probably invested in trunk thickness in response to water stress, adapted to flooding and thus gained a competitive advantage over other tree species. The thesis confirmed the assumption that Quercus robur is a resilient tree species to flooding and Tilia cordata showed higher resilience than expected.
Provedení detailního fytopatologického průzkumu a návrhy pro zlepšení stavu dřevin v Zoo Brno

The bachelor's thesis deals with the evaluation of the condition of woody plants, their vitality, health status, stability, operational safety and perspective. Furthermore, by proposing interventions to optimize their condition according to the methodology of the Nature and Landscape Protection Agency of the Czech Republic. A total of 200 trees were evaluated at the site - 11 small-leaved lindens (Tilia cordata Mill.), 11 hornbeam (Carpinus betulus L.) and 178 white oaks (Quercus petrea Liebl.). A field survey was carried out on these trees from March 2021 to March 2022. The results show that the trees on the site are not in good condition, their vitality and health are reduced. The presence of wood-destroying fungi such as Laetiporus sulphureus (Bull. Murrill) and Agrilus biguttatus (Fabricius, 1776) has also been discovered.

The bachelor thesis is focused on the topic: Use of instrumental methods to evaluate the condition of trees. The first part of the literature review describes the vitality of trees and explains why they are evaluated. Subsequently, the work deals with the division of individual methods and devices into visual and instrumental evaluation. These are then further categorized according to their principle of use.Totaly were evaluated four instrumental methods of physiological vitality and for biomechanical vitality six visual and eighteen instrumental method. In the next part, based on the description of these instrumental methods, an analysis of companies in the Czech Republic was created, which provide measurement and evaluation of expert opinions with these instruments.
